After you create the Single Template using the Layout Builder you need to assign it to the Portfolio Items as shown in the given screenshot. I have checked your custom layout and it seems it is already working. When viewing the live page, it is exactly used by the portfolio item.

As for the slider in your homepage, please check each of the Slide settings because right now, there is a slide with a padding of 10 pixels and there is also a padding of just 5 pixels.
