I’m not satisfied with the blog-like layout of Portfolio item pages, is there any way I can come up with my own layout? I’d like to have a full-width layout and mix of images and video gallery. I wonder if I can built my own layout with Cornerstone builder for that.

Basically, yes. You can create a blank page and use Cornerstone to add images linking to your portfolio items and you can also add a video gallery. This would require manual linking though.
If you want to auto-populate a page with portfolio items and want to have more design options, you will either need to use The Grid or the Essential Grid plugin. Both are bundled in X and Pro. Please see the links below.
