Is M4V necessary or will MP4 work?

If you’re referring to the Video shortcode, MP4 would suffice however, some browsers like Firefox does not fully support MP4 so you need to have different formats.
If that is not the case, please give us more details.

There’s virtually no difference between these two container formats. Both are QuickTime based, with one being slightly more catered to Apple’s platform than the other, and both offer quality-minded compression for the web. Most notably, both are used for the H.264 video codec, which is what you should be using for your videos (with a capped framerate and limited keyframes). MP4 also handles the MPEG-4 codec, but you will not want this for your video online, as it is centered more for physical distribution and not digital (meaning larger file sizes and slightly higher quality).
However, the concern above about Firefox is spot on… the same is also true with mp3 audio in many instances. The type of process used to package the file into these containers dictates whether the browser will support it… and with m4v, you cannot screw that part up (because it has only one use, and one codec.
In a nut shell… both will work, but use m4v. If this is for the intro slider (above the masthead as an example), I recommend using Compressor by Apple, and exporting 720p for Apple TV as your device preset to start out with before making any finer adjustments. This will give you optimum performance for the modern browser that loads it.
